Information on the Iowa State Mutator and other stocks from the Robertson laboratory.

In last year's News Letter, I indicated that I would be sorting through the stocks I had stored at ISU. This task has been completed and there are now two sources of the stocks that were retained, which are now available for distribution to interested researchers. One set of stocks was sent to the Maize Genetic Cooperation Stock Center at the University of Illinois, Urbana, Il. The second set of stocks has been retained at ISU. The Coop stocks are those I thought would be of more general interest to the maize genetic community, while the ISU stocks are those that I thought would be of more interest to Mutator connoisseurs.
Some item have been retained in both collections. Below are lists of the two sets of stocks. All stocks except those used as planting sources for, or produced in, the years 1989. 1990, and 1991 have been stored in our cold storage facilities. These facilities were not the most reliable and at times the stocks were in less than optional conditions for periods of a week or two until repairs could be made. Thus, it would be advisable to test a few kernels of each stock for germination before planting them in the field. The older the stocks the higher the likelihood of poor germination.

DEFINITIONS OF TERMS AND SYMBOLS.

Mutator (Mu) - A stock that has the potential for Mutator
activity.

MuDR - The autonomous element of the Mutator system.

Mu2 - Stocks resulting from the crossing of two active
Mutator lines, which have previously been propagated by outcrossing.

Mu3 - Stocks resulting from the crossing of an active
Mutator line with a Mu2 stock.

Mu4 - Stocks resulting from the crossing of two Mu2 lines.

Mu5 - Stocks resulting from the crossing of active
Mutator lines as follows: 1 X 4 = Mu X Mu4 or
2 X 3 = Mu2 X Mu3). Similarly for Mu6, Mu8, etc.

Outcrossed - Used when a Mutator stock or a Mutator-induced mutant is outcrossed one or more times to a Stand. or a Pl. Aleur. line or other genetic stocks.

Gene symbol-Mu - A Mutator-induced allele or event at the
locus the symbol represents.

Gene symbol-Mum - A mutable Mutator-induced allele at the
the locus the symbol represents.

Gene symbol-Mum stable - A stable derivative of a Mum allele.

Gene symbol-Mus - A Mutator-induced mutant that was non-mutable when first observed.

(1:1) - When associated with a genotype, indicates that the
particular Mutator-induced mutant segregated approximately 50% mutable to 50% stable mutant kernels. These stocks have the possibility of segregating for one or more MuDR elements.

(3:1) - When associated with a genotype, indicates that the particular Mutator-induced mutant segregated
approximately 75% mutable to 25% stable mutant
kernels. These stocks have the possibility of segregating two or more MuDR elements.

a1 sh2 segregant from (1:1) - One half of these stocks should carry one or more MuDR elements.

"Tested negative for Mutator activity." - Samples of kernels
from the ears of these stocks were sown and the resulting plants self-pollinated. None of the self-pollinated ears segregated for new mutants.

"Tested positive for Mutator activity." - Samples of kernels from the ears of these stocks were sown and the resulting plants self-pollinated. Some of the self-pollinated ears segregated for new mutants.

"Sibling plant(s) segregated for a new mutant" - Outcrossed ears from a family derived from the kernels of an active Mutator plant, which itself had been outcrossed. One or more of the sibling plants of that family segregated for a new mutant. (Note in these stocks and those in the previous definition have a high likelihood of having Mutator activity. However, some plants resulting from these crosses will have lost Mutator activity. Thus, to be sure an active plant has been selected for crossing it should be tested in some manner to determine that Mutator activity is retained. (See the paper in the News Letter entitled, "Some Thoughts on the
nature and Utilization of the Mutator System".)

Pl. Aleur - a purple aleurone stock homozygous for all alleles necessary for aleurone pigmentation.

Pl. (plus a genotype) - The given genotype is in a purple aleurone background as defined above.

Stand. - anyone of several different standard (F1 hybrid) lines.

homo. - Homozygous.

2 - Mutability class with only one or two reversion spots.

2 X 2 = 1 - Results observe when two Class 2 stocks are
crossed yielding progeny with only stable kernels (= Class 1).

Hi loss - A stock of an intensely mutable Mutator-induced mutant, which upon outcrossing has lost mutability in one generation, (i.e., the stock has become stable).
